Acetyl octapeptide-3. Topical peptide studied for SNARE-complex inhibition in wrinkle models.
Snap-8 (Acetyl Octapeptide-3) is an eight-amino-acid synthetic peptide built as an extension of Argireline (Acetyl Hexapeptide-3), with alanine and aspartate added to the original hexapeptide sequence. It was developed by the Spanish biotechnology company Lipotec (now part of Lubrizol Life Science) as a topical cosmeceutical for dynamic expression lines.
Mechanistically, Snap-8 mimics the N-terminal end of SNAP-25, the synaptosomal-associated protein that, together with syntaxin and VAMP, forms the SNARE complex at the presynaptic neuromuscular junction. SNARE assembly docks acetylcholine-loaded vesicles to the presynaptic membrane so they can fuse and release acetylcholine onto the muscle. By competitively occupying the SNAP-25 binding site, Snap-8 destabilises SNARE assembly, vesicle fusion falls, less acetylcholine is released, and the underlying micro-contraction is attenuated. This is the same molecular site botulinum toxin acts on, but the mechanism differs fundamentally: botulinum toxin enzymatically cleaves SNAP-25, producing paralysis lasting three to four months, while Snap-8 is a non-cleaving competitive modulator whose effect is transient and reverses on discontinuation. Topical application does not penetrate deeply enough to affect resting tone or motor function. Manufacturer head-to-head data report about 30% greater wrinkle-depth reduction than Argireline at equivalent concentration; no independent peer-reviewed trial exists.
